ChIPシークエンシング実験 (ChIP-seq) を成功させるためには、ターゲットタンパク質を正確に認識し、他のDNA結合タンパク質への非特異的な結合を示さない抗体を選ぶことが重要です。現在、CSTではリコンビナントラビットモノクローナル抗体をChIP-seqで実際に使用できるかを検証しています。なぜなら、ChIP-seqでは多様な遺伝子座に結合するターゲットタンパク質の広範な認識性能が必要とされるため、ChIP-qPCRで良いパフォーマンスを示す抗体がChIP-seqで使用できるとも限らないからです。私たちのChIP-seq推奨抗体は、SimpleChIP® Enzymatic Chromatin IP Kitのプロトコールと次世代シークエンサー (NGS) を用いて試験しています。

#13440 BRD4 (E2A7X) Rabbit mAb

ChIP was performed with cross-linked chromatin from 4 x 106 MV-4-11 cells and either 7 μg of BRD4 (E2A7X) Rabbit mAb #13440 or 7 μg of Competitor Polyclonal Antibody, using SimpleChIP® Plus Enzymatic Chromatin IP Kit (Magnetic Beads) #9005. DNA Libraries were prepared from 5 ng enriched ChIP DNA using NEBNext® Ultra II DNA Library Prep Kit for Illumina® and sequenced on the Illumina NextSeq. The CST recombinant rabbit monoclonal antibody provides higher signal and lower background than the Competitor Polyclonal Antibody in both whole genome analysis (Figure A) and localized gene analysis (Figure B).

#3418 CTCF (D31H2) XP® Rabbit mAb
